What is Chronic Fatigue Syndrome?

How I used to spend my days
"In the beginning I was a very fit and healthy person. I regularly competed in triathlons, 400km multi-day kayak races, participated in 500km 24 hours cycling events, ran up mountains and sometimes worked 80 hours a week.  I did whatever I wanted to do.

And then, over a period of time, a series of unfortunate events caused the 'wheels to fall off' my life. I contracted Ross River Fever, Glandular fever and some other mosquito-borne flavi-virus and this developed into chronic fatigue


Then I was exposed to a huge chemical leak which had to be cleaned up without any appropriate safety equipment - That’s when my atrial fibrillationbegan (an irregular and very rapid heartbeat).  Just to top things off, for whatever reason, I developed sensitivities to all sorts of food, like gluten and dairy products. Try avoiding those products; it's not easy.

And so for 15 years I’ve wandered in a wilderness of poor health and sometimes misery looking for some answers. In the end I could no longer work, and spent a lot of my time just lying in bed apparently looking like ‘death warmed up’ as they say. 


That was less than a year ago. Now I’m back on my bike, back in the garden, back into work and back into life, thanks to Cellgevity.  Today, I feel like I’m back from the dead! "

What is endometriosis?


Endometriosis is the abnormal growth of cells (endometrial cells) similar to those that form the inside or lining the tissue of the uterus, but in a location outside of the uterus. Endometrial cells are cells that are shed each month during menstruation. The cells of endometriosis attach themselves to tissue outside the uterus and are called endometriosis implants. These implants are most commonly found on the ovaries, the Fallopian tubes, outer surfaces of the uterus or intestines, and on the surface lining of the pelvic cavity. They can also be found in the vagina, cervix, and bladder, although less commonly than other locations in the pelvis. Rarely, endometriosis implants can occur outside the pelvis, on the liver, in old surgery scars, and even in or around the lung or brain. Endometrial implants, while they can cause problems, are benign (not cancerous).



Clinical studies have found that oxidative stress might play a role in the development and progression of endometriosis.

Markers of oxidative stress in follicular fluid of women with endometriosis and tubal infertility undergoing IVF.

Abstract

Oxidative stress and trace elements in the oocytes environment is explored in endometriosis and impact on in vitro fertilization (IVF) outcome assessed. Follicular fluid was aspirated at the time of oocyte retrieval from endometriosis (n=200) and tubal infertility (n=140) and the analytes measured using spectroscopy and HPLC. Increased concentration of reactive oxygen species (ROS), nitric oxide (NO), lipid peroxidation (LPO), iron, lead, cadmium and reduced levels of total antioxidant capacity (TAC), superoxide dismutase (SOD), catalase, glutathione peroxidase (GPx),glutathione reductase (GR), vitamins A, C, E, copper, zinc and selenium was observed compared to tubal infertility. Increased ROS and NO inendometriosis and tubal infertility associated with poor oocytes and embryo quality. Increased levels of ROS, NO, LPO, cadmium and lead were observed in women who did not become pregnant compared to women who did. Intrafollicular zinc levels were higher in women with endometriosiswho subsequently became pregnant following IVF.

Oxidative stress and antioxidants: exposure and impact on female fertility

Abstract

BACKGROUND Reproductive failure is a significant public health concern. Although relatively little is known about factors affecting fertility and early pregnancy loss, a growing body of literature suggests that environmental and lifestyle factors play an important role. There is sufficient evidence to hypothesize that diet, particularly its constituent antioxidants, and oxidative stress (OS) may influence the timing and maintenance of a viable pregnancy. We hypothesize that conditions leading to OS in the female affect time-to-pregnancy and early pregnancy loss.
METHODS We review the epidemiology of female infertility related to antioxidant defenses and oxidation and examine potential sources of OS from the ovarian germ cell through the stages of human pregnancy and pregnancy complications related to infertility. Articles were identified through a search of the PubMed database.
RESULTS Female OS is a likely mediator of conception and threshold levels for OS exist, dependent on anatomic location and stage of preconception.
CONCLUSIONS Prospective pregnancy studies with dietary assessment and collection of biological samples prior to conception with endpoints of time-to-pregnancy and early pregnancy loss are needed.

Abstract

BACKGROUND: Little is known about the aetiology of endometriosis; however, in the presence of oxidative stress, reactive oxygen species might increase growth and adhesion of endometrial cells in the peritoneal cavity, leading to endometriosis and infertility. Within a study investigating persistent organic compounds and endometriosis, the authors evaluated the association between oxidative stress and endometriosis. METHODS: Women aged 18–40 years who were undergoing laparoscopy were contacted to participate in the study (n=100); 84 were eligible and agreed to be interviewed; 78 provided blood specimens. Four markers of oxidative stress and antioxidant status were measured in serum for 61 women. Multiple imputation of missing data was used to generate values for the missing oxidative stress data. RESULTS: Thirty-two women had visually confirmed endometriosis at laparoscopy while 52 did not, including 22 undergoing tubal ligation and 30 with idiopathic infertility. There was a weak association between thiobarbituric acid-reactive substances (nmol/ml) and endometriosis, after adjusting for age, body mass index, current smoking, hormone use in the past 12 months, gravidity, serum vitamin E, serum estradiol, and total serum lipids (β=1.18; 95% CI–0.04, 2.39). CONCLUSIONS: These results suggest that oxidative stress might play a role in the development and progression of endometriosis, which should be evaluated in larger studies.

Gluten free and still in pain?

Colitis and Irritable Bowel Disease and the Role of Glutathione

What is colitis?

Colitis is inflammation of the inner lining of the colon and can be associated with diarrhea, abdominal pain,bloating, and blood in the stool. This inflammation may be due to a variety of reasons, including the following:

What is Irritable Bowel Syndrome?

Around one in five Australians experiences the unpleasant symptoms of irritable bowel syndrome (IBS) at some time. These include abdominal pain, mucus in the stools, and alternating diarrhoea and constipation. Other terms for irritable bowel syndrome include ‘spastic colon’ and ‘irritable colon’. It seems that people with IBS have sensitive bowels that are easily ‘upset’. More women than men are prone to IBS, and symptoms tend to first occur in early adulthood.

The cause is unknown, but environmental factors such as changes of routine, emotional stress, infection and diet can trigger an attack. Research has shown that the neurotransmitter serotonin may be important in the symptoms of IBS, by altering the function of nerve cells in the bowel and causing changes in pain sensation and bowel function.

Irritable bowel syndrome doesn’t cause lasting damage and doesn’t contribute to the development of serious bowel conditions, such as cancer or colitis.

Symptoms of IBS


Some of the more common signs of irritable bowel syndrome include:
  • Abdominal pain or cramping that is often relieved by passing wind or faeces
  • Alternating diarrhoea and constipation
  • A sensation that the bowels are not fully emptied after passing a motion
  • Abdominal bloating
  • Mucus present in the stools
  • Nausea.
None of these symptoms are exclusive to IBS. It is unusual for IBS to produce these symptoms, for the first time, after the age of 40.

Comparative efficacies of 2 cysteine prodrugs and a glutathione delivery agent in a colitis model

Oxidant-mediated injury plays an important role in the pathophysiology of inflammatory bowel disease (IBD). Recently, antioxidants were shown to modulate colitis in mice. In this study, the protective effects of L-cysteine and glutathione (GSH) prodrugs are further evaluated against progression of colitis in a murine model. ICR mice were fed compounds incorporated into chow as follows: Group (A) received chow supplemented with vehicle. Group (B) was provided 2-(RS)-n-propylthiazolidine-4(R)-carboxylic-acid (PTCA), a cysteine prodrug. Group (C) received D-ribose-L-cysteine (RibCys), another cysteine prodrug that releases L-cysteine. Group (D) was fed L-cysteine-glutathione mixed sulfide (CySSG), a ubiquitous GSH derivative present in mammalian cells. After 3 days, the animals were further provided with normal drinking water or water supplemented with dextran sodium sulfate (DSS). Mice administered DSS developed severe colitis and suffered weight loss. Colonic lesions significantly improved in animals treated with PTCA and RibCys and, to a lesser extent, with CySSG therapy. Hepatic GSH levels were depleted in colitis animals (control vs DSS, P < 0.001), and normalized with prodrug therapies (control vs treatments,P > 0.05). Protein expressions of serum amyloid A and inflammatory cytokines [interleukin (IL)-6, IL-12, tumor necrosis factor-alpha (TNF-α), osteopontin (OPN)] were significantly increased in colitis animals and improved with therapies. Immunohistochemistry and Western blot analyses showed significant upregulation of the macrophage-specific markers, COX-2 and CD68, which suggests macrophage activation and infiltration in the colonic lamina propria in colitis animals. These abnormalities were attenuated in prodrug-treated mice.
In conclusion, these data strongly support the novel action of the PTCA against colitis, which further supports a possible therapeutic application for IBD patients.
